About Thrombate III

Thrombate III is the only therapy approved by the FDA specifically for the treatment of hereditary antithrombin (AT) deficiency. Thrombate III has no known contraindications.

Thrombate III is a sterile, nonpyrogenic, stable, lyophilized preparation of purified human antithrombin.1

Thrombate III is indicated to treat individuals with hereditary AT deficiency in connection with surgical or obstetrical procedures or when they suffer from thromboembolism.1

Antithrombin is the most important serine protease inhibitor of coagulation and plays a critical role in maintaining normal hemostasis.2

Thrombate III works just like native antithrombin. It:

Binds irreversibly to both factor Xa and thrombin (factor IIa)
The anticoagulation activity of AT is increased ~1,000-fold when it binds to heparin
Heparin has no anticoagulation activity without antithrombin
Prevents factor Xa from transforming prothrombin into thrombin
Prevents thrombin from activating platelets and participating in the conversion of fibrinogen into crosslinked fibrin polymer
Reduces the amount of fibrin available for clot formation

Thrombate III has a half-life in the circulation comparable to native AT1:
2.5 days based on immunological assays
3.8 days based on functional assays

Thrombate III is convenient to use.
  Ready to infuse in minutes
Should be refrigerated (2-8°C; 36-46°F) in the pharmacy, the operating room, or the blood bank1
Available in 500 IU single use vials—with Sterile Water for Injection, a sterile double-ended transfer needle, a filter needle, and dosing instructions included with each vial1

Important Safety Information
Thrombate III therapy has demonstrated a low occurrence of side effects. In clinical studies with Thrombate III, the most common side effects were dizziness, chest tightness, nausea, and a foul taste in the mouth. As with all plasma-derived therapeutics, the potential to transmit infectious agents, such as viruses, cannot be totally eliminated. Individuals who receive infusions of blood or blood plasma may develop signs and/or symptoms of some viral infections, particularly hepatitis C.
